STEP 2. SYNTHESIS OF (E)-1-ETHOXY-2- (1, 8-NAPHTHYRIDIN-2- YL) ethanol. To the product from step 1, (81.5 G, 0.57 mol) in anhydrous THF (1.9 L) at – 40 C under Ar gas was added lithium bis (trimethylsilyl) amide (1 M in THF, 1.2 L, 1.2 MOL). After stirring for 30 min at-40 C, DIETHYLCARBONATE (72.5 mL, 0.60 mol) was added. The temperature of the reaction mixture was warmed up to 0 C and stirred for 2 h. The reaction mixture was quenched into saturated aq. NH4CI (700 mL) and the THF removed under reduced pressure. The resulting mixture was extracted with EtOAc (3 x 700 mL). The organic layers were combined, washed with brine, dried over NA2SO4, and concentrated under reduced pressure. The resulting residue was purified by flash column chromatography using 50percent EtOAc/hexane to give a yellow solid (81.2 g, 0.38 mol, 66percent).APOS;H NMR (400 MHz, DMSO-d6) 8 1.22 (t, 3H), 4.11 (q, 2H), 4.89 (s, 1H), 6.78 (d, 1H), 7.15 (dd, 1H), 7.47 (d, 1H), 7.80 (d, 1H), 8.36 (d, 1H), 11.8 (bs, 1H). LC-MS (MH+) = 217.

The reaction mixture was cooled to room temperature, and added dropwise to a mixture of ethyl acetate (30 mL), water (30 mL), and sodium carbonate (9.57 g) over a period of 1 hour in an ice bath. Water (10 mL) was added thereto, and sodium carbonate was added thereto, followed by adjusting the pH of the resultant product to 8.3. The resultant product was stirred at room temperature for 10 minutes, and ethyl acetate (270 mL) and water (200 mL) were added thereto. The organic layer was collected by separation, and the aqueous layer was extracted two times with ethyl acetate (200 mL). The organic layer and the extraction liquid were combined, the resultant product was dried over anhydrous sodium sulfate, and the solvent was distilled off under reduced pressure, thereby obtaining 2-chloro-1,5-naphthyridine (2.86 g) as a pale yellow solid. MS m/z (M+H): 165.

The solvent was removed under vacuum and the residue waspurified by column chromatography eluting with DCM/MeOH (20/1, R1= 0.4) to give 350mg of tertbutyl N-(1 ,8-naphthyridin-4-yl)carbamate as a brown solid.A suspension of 280mg (1.14 mmol) of tert-butylN-(1,8-naphthyridin-4-yl)carbamate and 164 mg (0.12 mmol) of NaH in 2 ml. of THF was stirred at 40C for 16 h. A solution of 0.04 g (0.13 mmol) of 4- bromo-6-(bromomethyl)isoquinoline in 2 mL of THF was added thereto at 3 5C. The mixture was stirredat 35C for additional 0.5 h. The reaction was quenched with saturated aqueous NH4C1 solution. The mixture was diluted with EtOAc and washed with brine. The ethyl acetate solution is washed with 2 N NAOH and water, dried over magnesium sulphate, then concentrated to afford the title compound. MS: APCI 240.0 MH+.

A stirred solution of 2-methyl-l,8-naphthyridine (57.5 g, 399 mmol) (available from Manchester Organics) and ( ?)-fe/ -butyl 3-(iodomethyl)pyrrolidine-l-carboxylate (124.2 g, 399 mmol) (Intermediate 1) in THF (1 L) was cooled to 0 ¡ãC and treated under nitrogen with a solution of lithium bis(trimethylsilyl)amide in THF (1M, 399 ml_, 399 mmol) over 20 min and the reaction mixture was stirred at 0 ¡ãC for 3 h. The reaction was quenched with saturated ammonium chloride solution (500 ml.) and water (500 ml.) and ethyl acetate (1 L) was added. The layers were separated and the aqueous phase was extracted with further ethyl acetate (1 L). The combined organic layers were dried (MgS04), filtered and evaporated in vacuo. The residual brown oil (162 g) was purified by chromatography on a silica cartridge (750 g) eluting with a gradient of 0 – 100 percent [ethyl acetate in (5percent MeOH – 95 percent ethyl acetate)] over 8 column volumes. The appropriate fractions were combined and evaporated in vacuo to give the title compound (46.65 g, 36percent) as an orange solid: LCMS (System A) RT = 0.99 min, 97percent, ES+ve m/z 32S (M+H)+, [a]D20 = + 22 (c 1.00 in EtOH).

The organic layer was collected by separation, washed with a saturated sodium chloride aqueous solution, and dried over anhydrous sodium sulfate, and the solvent was distilled off under reduced pressure. The obtained residue was purified by silica gel column chromatography (ethyl acetate-hexane), thereby obtaining tert-butyl 3-(4-(6-chloro-1,5-naphthyridin-3-yl)-1H-pyrazol-1-yl)azetidine-1-carboxylate (63 mg) as a white solid. A SOLUTION OF AMINE 31 (612 MG, 1.57 MMOL) and triethylamine (0.7 mL, 5.0 MMOL) in ACETONITRILE (4 mL) was treated with 7-CHLORO-1-CYCLOPROPYL-6-FLUORO-4-OXO-1, 4-dihydro-naphthpyridine- 3-carboxylic acid (222 mg, 0.787 MMOL) under nitrogen and the reaction mixture was allowed to stir for 12 h. The solid was collected, resuspended in methanol (5 mL) and the reaction mixture was treated with hydrazine (1 mL). After 5 min, the reaction mixture was warmed to reflux and the resulting mixture was allowed to stir for 1 h. The reaction mixture was concentrated in vacuo, diluted with water and the solids were collected by filtration. The off white product was washed with water (3 x 20 mL), allowed to dry overnight to afford the title compound 1 (40.4 mg, 13%). MS 391 (M+H).

With ice cooling and stirring, to the solution was added 600 mg of potassium nitrate in portions. The solution was stirred at room temperature for 30 minutes to complete nitration. Then the reaction solution was poured into a stirred mixture of 150 ml of chloroform and 100 ml of ice water. After stirring at room temperature for 15 minutes, the reaction solution separated. The chloroform layer was dried over anhydrous magnesium sulfate and then concentrated in vacua. The precipitated crystals were dispersed in ethanol, collected by filtration and washed with ethanol and then with diisopropyl ether to give 2.08 g of the title compound.
