Fluoroquinolones are synthetic antibacterial agents that stabilize the ternary complex of prokaryotic topoisomerase II enzymes (gyrase and Topo IV), leading to extensive DNA fragmentation and bacteria death. Despite the similar structural folds within the critical regions of prokaryotic and eukaryotic topoisomerases, clinically relevant fluoroquinolones display a remarkable selectivity for prokaryotic topoisomerase II, with excellent safety records in humans. Typical agents that target human topoisomerases (such as etoposide, doxorubicin and mitoxantrone) are associated with significant toxicities and secondary malignancies, whereas clinically relevant fluoroquinolones are not known to exhibit such propensities. Although many fluoroquinolones have been shown to display topoisomerase-independent antiproliferative effects against various human cancer cells, those that are significantly active against eukaryotic topoisomerase show the same DNA damaging properties as other topoisomerase poisons. Empirical models also show that fluoroquinolones mediate some unique immunomodulatory activities of suppressing pro-inflammatory cytokines and super-inducing interleukin-2. This article reviews the extended roles of fluoroquinolones and their prospects as lead for the unmet needs of small and safe multimodal-targeting drug scaffolds.

Twelve new complexes, namely [Ag-2(pyNP)(2)(NO3)(2)] (2), [Hg(pyNP)X-2](X = Cl(3), Br(4), l(5), CN(6), SCN(7, 8)), [Pb(pyNP)(2)(NO3)(2)] (9), [Pb (pyNP)(NO3)(2)](2) (10), [Cu(pyNP)Cl-2(H2O)] (11), [Cu(pyNP)(2)(H2O)[Hg-2(CN)(4)Cl-2]-H2O (12) and [Cu(pyNP) (H2O)(2)(mu-CN)Hg-2(CN)(3)Cl-2]-H2O (13), have been synthesized and characterized by single crystal X-ray diffraction, Raman, FTIR and electronic spectroscopies. Highly Efficient Electron-Transporting/Injecting and Thermally Stable Naphthyridines for Organic Electrophosphorescent Devices

A series of 1,8-naphthyridine derivatives is synthesized and their electron-transporting/injecting (ET/EI) properties are investigated via a multilayered electrophosphorescent organic light-emitting device (OLED) using fac-tris(2-phenylpyridine)iridium [Ir(ppy)3] as a green phosphorescent emitter doped into a 4,4-N,N-dicarbazolebiphenyl (CBP) host with 4,4-bis[N-(1-naphthyl)-N-phenylamino]biphenyl (a-NPD) as the hole-transporting layer, and poly(arylene ether sulfone) containing tetraphenylbenzidine (TPDPES) doped with tris(4-bromophenyl)ammonium hexachloroantimonate (TBPAH) as the hole-injecting layer. The turn-on voltage of the device is 2.5 V using 2,7-bis[3-(2-phenyl)-1,8-naphthyridinyl]-9,9-dimethylfluorene (DNPF), lower than that of 3.0 V for the device using a conventional ET material. The maximum current efficiency (CE) and power efficiency (PE) of the DNPF device are much higher than those of a conventional device. With the aid of a hole-blocking (HB) and exciton-blocking layer of bathocuproine (BCP), 13.213.7% of the maximum external quantum efficiency (EQE) and a maximum PE of 50.254.5 lm W1 are obtained using the naphthyridine derivatives; these values are comparable with or even higher than the 13.6% for conventional ET material. The naphthyridine derivatives show high thermal stabilities, glass-transition temperatures much higher than that of aluminum(III) bis(2-methyl-8-quinolinato)-4-phenylphenolate (BAlq), and decomposition temperatures of 510518 degrees C, comparable to or even higher than those of Alq3.

Synthesis and biological evaluation of 3-(1,3,4-oxadiazol-2-yl)-1,8-naphthyridin-4(1H)-ones as cisplatin sensitizers

A series of novel 3-(1,3,4-oxadiazol-2-yl)-1,8-naphthyridin-4(1H)-one derivatives were synthesized and their anti-cancer as well as cisplatin sensitization activities were evaluated. Among them, compounds 6e and 6h exhibited significant cisplatin sensitization activity against HCT116. Hoechst staining and annexin V-FITC/PI dual-labeling studies demonstrated that the combination of 6e/6h and cisplatin can induce tumour cell apoptosis. Western blot showed that the expression of ATR downstream protein, CHK1, decreased in 6e + cisplatin and 6h + cisplatin groups compared with that in the test compound and cisplatin group. Furthermore, docking of 6e/6h into the ATR structure active site revealed that the N1 and N8 atoms in the naphthyridine ring and the hybrid atom in the oxadiazole ring are involved in hydrogen bonding with Val170, Glu168 and Tyr155. Additionally, the naphthyridine ring is also involved in – stacking with Trp169. Accordingly, compounds 6e and 6h can be expected to be potential cisplatin sensitizers that can participate in HCT116 cancer therapy.
