Synthesis and DNA binding of 6-(alkylamino)indolo[1,2-b][2,7]naphthyridine-5,12-quinones

We describe the synthesis of eight novel putative mono- and bis-DNA intercalators from a common precursor, 6-bromoindolo[1,2-b][2,7]naphthyridine-5,12-dione. Of these new indoloquinones, our data indicate that two are most likely DNA mono-intercalators, but weaker than ethidium bromide, and two others are DNA bis-intercalators. Our indoloquinones are inactive against mammalian topoisomerase II. [GRAPHICS] .

SYNTHESIS AND IN VITRO ANTIPROLIFERATIVE SCREENING OF NEW 2,7-NAPHTHYRIDINE-3-CARBOXYLIC ACID HYDRAZIDE DERIVATIVES

The new pyrrolo[3,4-c]pyridines and 2,7-naphthyridine derivatives have been synthesized. 4-Hydroxy-8-methyl-l-oxo-6-phenyl-1,2-dihydro-2,7-naphthyridine-3-carboxylic acid hydrazide (4) was the key intermediate for the synthesis of the novel derivatives of various chemical structures: Schiff bases, 1,3,4-oxadiazoles, pyrazoles, carbohydrazides, semi- and thiosemicarbazides. The structures of these new compounds were confirmed by elemental analysis and IR, NMR and MS spectra. The antitumor activities of the obtained derivatives were examined. Eight of the twenty one newly synthesized compounds were qualified by the NCI (Bethesda, MD, USA) for in vitro screening against 60 different human tumor cell lines. The most active proved to be the Schiff bases.

A 2, 7-naphthyridine-based fluorescent turn-on probe for detection of biothiols in vitro and in vivo

A 2,7-naphthyridine derivative (AND-OH) was used as a fluorophore in the development of a novel fluorescent turn-on probe (AND-DNBS) by introducing a 2,4-dinitrobenzenesulfonate functionality. This fluorescent probe was successfully used for sensing glutathione (GSH) in aqueous media and showed favorable properties such as a rapid response time (45 s) and a significant Stokes shift (227 nm). The characteristics of the developed probe are better than those of another probe, namely ND-DNBS, which was also synthesized by our group and most other previously reported fluorescent probes for GSH. The AND-DNBS probe was able to detect GSH via a red emission in living A549 cells and zebrafishes. These results show that AND-DNBS has potential applications as an excellent probe for detecting GSH in various environments.

A Novel Ruthenium(II) Polypyridyl Complex Bearing 1,8-Naphthyridine as a High Selectivity and Sensitivity Fluorescent Chemosensor for Cu2+ and Fe3+ Ions

A novel ruthenium(II) polypyridyl complex bearing 1,8-naphthyridine was successfully designed and synthesized. This complex was fully characterized by EI-HRMS, NMR, and elemental analyses. The recognition properties of the complex for various metal ions were investigated. The results suggested that the complex displayed high selectivity and sensitivity for Cu2+ and Fe3+ ions with good anti-interference in the CH3CN/H2O (1:1, v/v) solution. The fluorescent chemosensor showed obvious fluorescence quenching when the Cu2+ and Fe3+ ions were added. The detection limits of Cu2+ and Fe3+ were 39.9 nmol/L and 6.68 nmol/L, respectively. This study suggested that this Ru(II) polypyridyl complex can be used as a high selectivity and sensitivity fluorescent chemosensor for Cu2+ and Fe3+ ions.

Catalyst-free Glycerol Promoted Green Synthesis of 2-amino-1,8-naphthyridine-3-carbonitriles and 2-amino-3-quinolinecarbonitriles

Influence of substituent modifications on the binding of 2-amino-1,8-naphthyridines to cytosine opposite an AP site in DNA duplexes: thermodynamic characterization

Here, we report on a significant effect of substitutions on the binding affinity of a series of 2-amino-1,8-naphthyridines, i.e., 2-amino-1,8-naphthyridine (AND), 2-amino-7-methyl-1,8-naphthyridine (AMND), 2-amino-5,7-dimethyl-1,8-naphthyridine (ADMND) and 2-amino-5,6,7-trimethyl-1,8-naphthyridine (ATMND), all of which can bind to cytosine opposite an AP site in DNA duplexes. Fluorescence titration experiments show that the binding affinity for cytosine is effectively enhanced by the introduction of methyl groups to the naphthyridine ring, and the 1:1 binding constant (10(6) M-1) follows in the order of AND (0.30) AMND (2.7) ADMND (6.1) ATMND (19) in solutions containing 110 mM Na (pH 7.0, at 20C). The thermodynamic parameters obtained by isothermal titration calorimetry experiments indicate that the introduction of methyl groups effectively reduces the loss of binding entropy, which is indeed responsible for the increase in the binding affinity. The heat capacity change (C-p), as determined from temperature dependence of the binding enthalpy, is found to be significantly different between AND (161 cal/mol K) and ATMND (217 cal/mol K). The hydrophobic contribution appears to be a key force to explain the observed effect of substitutions on the binding affinity when the observed binding free energy (G(obs)) is dissected into its component terms.

Catalytic Reduction of Nitroarenes by Dipalladium Complexes: Synergistic Effect

The direct reaction between 2,7-bis(2-pyridinyl)-1,8-naphthyridine (bpnp) and Pd(CH3COO)(2) in CF3COOH yields the new dinuclear palladium(II) complex [Pd-2(bpnp)(mu-OH)(CF3CO2)(2)](CF3CO2) (1). Similarly, substitution of Pd(CH3CN)(4)(BF4)(2) with bpnp in DMF gives [Pd-2(bpnp)(mu-OH)(DMF)(2)](BF4)(3) (2). Treatment of 1 or 2 with Cl- readily provide the chloro-substituted species [Pd(bpnp)(mu-OH)(Cl)(2)](+). All complexes were characterized by spectroscopic methods, and the structure of 2 was further confirmed by X-ray crystallography. Complex 1 is an efficient catalyst for the reduction of aromatic nitro compounds leading to the corresponding aniline derivatives under atmospheric pressure of hydrogen at 50 degrees C. The mechanistic pathway of the catalysis is investigated. From the reaction pathway, it is suggested that a facile condensation of nitroso and hydroxylamine intermediates is enabled by the dipalladium system and the desired transformation proceeds smoothly under mild reaction conditions to yield the reduced product.

1,8,10-Trisubstituted anthracenyl hydrocarbons: Towards versatile scaffolds for multiple-H-bonded recognition arrays

In this work, we describe the synthesis of 1,8,10-trisubstituted anthracenyl scaffolds that, bearing boronic acid functionalities, can act as multiple H-bonding donor systems. The trisubstituted anthracenyl derivatives are synthesized following two main synthetic pathways. Whereas in the first approach trisubstituted anthracenyl derivatives are prepared through the regioselective addition of the relevant organomagnesium nucleophile to 1,8-dichloroanthraquinone, in the second avenue a triflate-bearing anthracene is prepared by reduction of the anthraquinone into the anthrone precursor and functionalized through metal-catalysed cross-coupling reactions. Complementary studies of the Na2S2O4-mediated reduction of 1,8-dichloroanthraquinone allowed to shed further light on the possible mechanism of formation of the anthrone precursor, suggesting the presence of a cis-diol intermediate undergoing antiperiplanar elimination. Solid-state X-ray diffraction investigations of the bisboronic acids show that the molecules self-assemble into dimers through the formation of four H-1-bonds established between the anti-syn conformers of the boronic acid moieties. H-1-NMR titrations between bisboronic acids and tetra H-bond acceptor, diisoquinolino-naphthyridine, showed a significant shift of the -B(OH)(2) proton resonances, suggesting the presence of H-bonding interactions between both molecules. Discovery of 7-Aryl-Substituted(1,5-Naphthyridin-4-yl) ureas as Aurora Kinase Inhibitors

As part of our research projects to identify new chemical entities of biological interest, we developed a synthetic approach and the biological evaluation of (7-aryl-1,5-naphthyridin-4-yl)ureas as a novel class of Aurora kinase inhibitors for the treatment of malignant diseases based on pathological cell proliferation. 1,5-Naphthyridine derivatives showed excellent inhibitory activities toward Aurora kinases A and B, and the most active compound, 1-cyclopropyl-3-[7-(1-methyl-1H-pyrazol-4-yl)-1,5-naphthyridin-4-yl]urea (49), displayed IC50 values of 13 and 107nM against Aurora kinases A and B, respectively. In addition, the selectivity toward a panel of seven cancer-related protein kinases was highlighted. In vitro ADME properties were also determined in order to rationalize the difficulties in correlating antiproliferative activity with Aurora kinase inhibition. Finally, the good safety profile of these compounds imparts promising potential for their further development as anticancer agents.

Reversible Intramolecular Cyclization in Ruthenium Complexes Induced by Ligand-centered One-electron Transfer on Bidentate Naphthyridine: An Important Intermediate for Both Metal- and Organo-Hydride Species
