Naphthyridine-based lanthanide complexes worked as magnetic resonance imaging contrast for guanosine 5 ‘-monophosphate in vivo

New lanthanide complex Gd-ANAMD containing 2-amino-7-methyl-1,8-naphthyridine was achieved for selective magnetic resonance imaging towards guanosine 5′-monophosphate over other ribonucleotide polyphosphates in aqueous media and in vivo. The formation of strong multi-hydrogen bonds between naphthyridine and guanosine made the phosphate in guanosine 5’-monophosphate positioned on a suitable site to coordinate with the lanthanide ion. The substitution of the coordination naphthyridine by the phosphate oxygen atoms caused obvious relaxivity decrease. The negligible cytotoxicity and appropriate blood circulation time of Gd-ANAMD allow potential application of Magnetic Resonance Imaging in vivo. H-1 NMR confirmed that the selectivity of these lanthanide complexes towards guanosine was attributed to the formation of hydrogen bonds between the guanine moeity and the naphthyridine. The fluorescence detection and lifetime measurement of Tb-ANAMD and Eu-ANAMD suggested that the decrease of the relaxivity is not attributed to the change of the q value, but caused by the prolonging of the residence lifetime of inner-sphere water. A One-Pot Diazotation-Fluorodediazoniation Reaction and Fluorine Gas for the Production of Fluoronaphthyridines

Several synthetic routes to 7-fluoro-2-methoxy-8-methyl-1,5-naphthyridine (1) are presented, and their suitability for scale-up is discussed. The way of introducing the fluorine atom is crucial. Early routes start from commercially available fluorinated building blocks or employ F+ reagents like Select Fluor and delivered up to 70 kg of 7-fluoro-2-methoxy-1,5-naphthyridine (18). To prepare for larger scales, the focus turned to the use of HF or elemental fluorine, both one of the cheapest sources of fluorine. The first method, a one-pot diazotation-fluorodediazoniation with 6-methoxy-1,5-naphthyridin-3-amine (9) in HF gave the fluorinated naphthyridine 18 in high yield and purity without isolation of the unstable diazonium salt, the latter being a severe drawback of the related Balz-Schiemann protocol. The second method relies on the use of fluorine gas for a surprisingly selective ortho-fluorination of 6-methoxy-1,5-naphthyridin-4-ol (10).

Targeting Expanded Repeats by Small Molecules in Repeat Expansion Disorders

Recent technological advancements in genetic analysis have allowed for the consecutive discovery and elucidation of repeat expansion disorders: diseases caused by the abnormal expansion of repeat sequences in the genome. Many of these repeat expansion disorders are neurodegenerative movement disorders. Radical cures for these disorders have yet to be established. Although conventional treatments for repeat expansion disorders have mainly targeted the abnormal mRNA and proteins encoded by the affected genes, therapeutic approaches targeting repeat DNA, the root cause of repeat disorders, is also being explored in current research. In particular, a small molecule has been found that binds to abnormally expanded CAG repeats, the cause of Huntington’s disease, and shortens them. Such small molecules targeting nucleic acids are expected to be developed into groundbreaking therapeutic drugs capable of ameliorating the symptoms of repeat expansion disorders and preventing their onset. Hydrogen Transfer-Mediated Multicomponent Reaction for Direct Synthesis of Quinazolines by a Naphthyridine-Based Iridium Catalyst

Selective linkage of renewable alcohols and ammonia into functional products would not only eliminate the prepreparation steps to generate active amino agents but also help in the conservation of our finite fossil carbon resources and contribute to the reduction of CO2 emission. Herein the development of a novel 2-(4-methoxyphenyl)-1,8-naphthyridine-based iridium (III) complex is reported, which exhibits excellent catalytic performance toward a new hydrogen transfer-mediated annulation reaction of 2-nitrobenzylic alcohols with alcohols and ammonia. The catalytic transformation proceeds with the striking features of good substrate and functional group compatibility, high step and atom efficiency, no need for additional reductants, and liberation of H2O as the sole by-product, which endows a new platform for direct access to valuable quinazolines. Mechanistic investigations suggest that the non-coordinated N-atom in the ligand serves as a side arm to significantly promote the condensation process by hydrogen bonding.

Enhancement of affinity in molecular recognition via hydrogen bonds by POSS-core dendrimer and its application for selective complex formation between guanosine triphosphate and 1,8-naphthyridine derivatives

We report that a polyhedral oligomeric silsesquioxane (POSS) core in a dendrimer can enhance the affinity of the molecular recognition via hydrogen bonds between 1,8-naphthyridine and guanosine nucleotides. The complexation of the naphthyridine ligands with a series of guanosine nucleotides was investigated, and it is shown that the POSS core should play a significant role in the stabilization of the complexes via hydrogen bonds. Finally, we demonstrate that the 1,8-naphthyridine ligand can selectively recognize guanosine triphosphate by assisting with the POSS-core dendrimer.

Tailor-made naphthyridines: Self-assembling multiple hydrogen-bonded supramolecular architectures from dimer to helix

Stepwise changes of functional oxo and amino groups in 1,8-naphthyridines to modify the supramolecular architecture have been carried out. The first example of a naphthyridine helix has been found and its structure established by X-ray crystallography. The design is based on hydroxy and amido tautomeric naphthyridines which crystallize in dimers or catemers, one of them attaining helicity. The most stable tautomer present in all the compounds discussed in this paper, as well as the formation of hydrogen-bonded dimers or catemers, was established by X-ray crystallography and rationalized with theoretical calculations. Supramolecular salts of 5,7-dimethy1-1,8-naphthyridine-2-amine and acids through classical H-Bonds and other intermolecular interactions

This article demonstrates 5,7-dimethyl-1,8-naphthyridine-2-amine based organic salts formation of crystalline solids 1-9, in which the acidic units have been integrated via a variety of non-covalent bonds. Addition of equivalents of the acidic units to the solution of 5,7-dimethyl-1,8-naphthyridine-2-amine generates the single protonated species which direct the anions. The nine compounds crystallize as their salts with the acidic H transferred to the aromatic N of the 5,7-dimethy1-1,8-naphthyridine-2-amine. In 2, 3, 4, and 7 the methyl substituted N-rings were protonated, while others were protonated at the NH2 substituted N-ring. All salts have been characterized via IR, mp, EA and XRD technique. The major driving force for the salt formation is attributed to the classical H-bonds from 5,7-dimethyl-1,8-naphthyridine-2-amine and the acids. Other extensive intermolecular interactions also play great functions in space association of the molecular assemblies in the relevant crystals. The common R-2(2)(8) graph set has been observed in all salts due to the H-bonds and other intermolecular interactions, except 2, 4, and 6. For the synergistic interactions of the various non-covalent bonds, all salts displayed 3D structures. (C) 2017 Elsevier B.V. All rights reserved.
