Studying quadruply hydrogen bonding (QHB) module interactions in materials matrices presents a significant challenge because a wide variety of non-covalent interactions may be relevant. Here we introduce a method of surface modification with DeUG (7-deazaguanine urea), DAN (2,7-diamido-1,8-naphthyridine) and UPy (2-ureido-4[1H]-pyrimidone) modules to form self-assembled monolayers (SAMs) on a glass surface. The QHB interactions under mechanical stress were investigated by measuring adhesion force using PS-DAN (DAN modified polystyrene), PBMA-DeUG (DeUG modified poly butyl methacrylate) and PBA-UPy (UPy modified poly butyl acrylate) as adhesion promoters. A mechanical lap-shear test was used to evaluate the fracture resistance of QHB heterocomplexes. The maximum load at fail showed that QHB interaction contributed significantly (72%) to overall adhesion. For the QHB modified glass surface, using a polymer modified with its complementary QHB partner greatly facilitated their pairing efficiency, up to 40% for DAN-DeUG. A general method from which single pair ruptures force of QHB modules could be obtained using thermodynamic data obtained from solution chemistry was proposed. Using this method, the single pair rupture force for UPy-UPy was measured as 160 pN, and the single pair rupture force for DAN-DeUG was obtained as 193 pN.

1,8-Naphthyridine derivatives related to 17 (ITH4012), a neuroprotective compound reported by our research group, have been synthesized. In general, they have shown better inhibition of acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) than most tacrine derivatives previously synthesized in our laboratory. This compound also exhibited neuroprotection against neurotoxicity elicited by oxygen and glucose deprivation in hippocampal slices. Because these stressors caused neuronal damage related to physiopathological hallmarks found in the brain of Alzheimer’s disease (AD) patients, we conclude that compound 14 deserves further in vivo studies in AD models to test its therapeutic potential in this disease.

A simple and convenient sequential one-pot synthesis of 1,2,3,4-tetrahydrobenzo[b][1,6] naphthyridines has been developed. In the present work three chromeno naphthyridine derivatives (CNs) have been synthesized using one pot multicomponent reaction of 2, 4-hydroxyacetophenone, aromatic aldehydes and malononitrile in water using silica gel as a catalyst. The simulated LDse values for the synthesized compounds are 340 mg/kg (CN-1), 1500 mg/kg (CN-2) and 800 mg/kg (CN-3) which suggest their non-toxic and eco-friendly nature The performance all the three compounds as corrosion inhibitors in 15% HCl on N80 has been analyzed via weight loss, electrochemical, surface characterization, and density functional theory (DFT) methods. Chromeno naphthyridine (CN-1) shows maximum inhibition efficiency of 98.3% at only 300 mg/L The measurement of EIS reveals the process of inhibition of corrosion is governed by control of charge transfer. The mixed nature of CNs adsorption is suggested by results of PDP. Langmuir model of adsorption shows the perfect fitting. Adsorption of CNs onto the metal surface was supported by MR. AFM, and UV-vis. Computational modelling using DFT data reveals that corrosion inhibition takes place by adsorption of both neutral and protonated forms on steel surface. Disruption of the hypothalamic-pituitary-adrenal axis is an established finding in patients with anxiety and/or depression. Chronic corticosterone administration in animals has been proposed as a model for the study of these stress-related disorders and the antidepressant action. Alterations of the central noradrenergic system and specifically of inhibitory alpha(2)-adrenoceptors seem to be part of the pathophysiology of depression and contribute to the antidepressant activity. The present study evaluates in male rats the effect of chronic corticosterone treatment during 35 days (16-20 mg kg(-1) day(-1)) on the sensitivity of alpha(2)-adrenoceptors expressed in the somatodendritic and terminal noradrenergic areas locus coeruleus (LC) and prefrontal cortex (PFC), respectively. Further, the effect of chronic fluoxetine treatment (5 mg kg(-1), i.p., since the 15th day) on the sensitivity of alpha(2)-adrenoceptors was examined under control conditions and in corticosterone-treated rats. The alpha(2)-adrenoceptor functionality was analysed in vitro by agonist-mediated [S-35]GTP gamma S binding stimulation and in vivo through the modulation of noradrenaline (NA) release evaluated by dual-probe microdialysis. The concentration-effect curves of the [S-35]GTP gamma S binding stimulation by the agonist UK14304 (5-bromo-N-(4,5-dihydro-1H-imidazol-2-yl)-6-quinoxalinamine) demonstrated a desensitization of cortical alpha(2)-adrenoceptors induced by corticosterone (-logEC(50) = 6.7 +/- 0.2 vs 8.2 +/- 0.3 in controls) that was reverted by fluoxetine treatment (-logEC(50) = 7.5 +/- 0.3). Local administration of the alpha(2)-adrenoceptor antagonist RS79948 ((8aR,12aS,13aS)-5,8,8a,9,10,11,12,12a,13,13a-decahydro-3-methoxy-12-(ethylsulfonyI)-6H-isoquino [2,1 -g] [1,6]naphthyridine) (0.1-100 mu mol L-1) into the LC induced a concentration-dependent NA increase in the PFC of the control group (E-max = 191 +/- 30%) but non-significant effect was observed in corticosterone-treated rats (E-max = 133 +/- 46%), reflecting a desensitization of a 2 -adrenoceptors that control the firing of noradrenergic neurons. Fluoxetine treatment did not alter the corticosterone-induced desensitization in this area (E-max = 136 +/- 19%). No effect of fluoxetine on alpha(2)-adrenoceptor functionality was observed in control animals (E-max = 223 +/- 30%). In PFC, the local administration of RS79948 increased NA in controls (E-max = 226 +/- 27%) without effect in the corticosterone group (E-max = 115 +/- 26%), suggesting a corticosterone-induced desensitization of terminal alpha(2)-adrenoceptors. Fluoxetine administration prevented the desensitization induced by corticosterone in the PFC (E-max = 233 +/- 33%) whereas desensitized alpha(2)-adrenoceptors in control animals (E-max = -24 +/- 10%). These data indicate that chronic corticosterone increases noradrenergic activity by acting at different alpha(2)-adrenoceptor subpopulations. Treatment with the antidepressant fluoxetine seems to counteract these changes by acting mainly on presynaptic alpha(2)-adrenoceptors expressed in terminal areas.
