A series of novel 2,7-dimethyl-1,8-naphthyridine derivatives substituted with Mannich bases 2a-d, N-beta-glycosides 6a-e, 7a-e, Schiff’s bases 8a-c, pyrazolone 9, and S-alkylated derivatives 10a-c have been synthesized in good yields starting from 4-hydroxy-2,7-dimethyl-1,8-naphthyridine 1 through multi-step synthesis. The newly synthesized title compounds were evaluated for their HepG2 cell growth inhibition, the results revealed that all the tested compounds process inhibitory effects on the growth of HepG2 liver cancer cells. Compound 8b showed the highest inhibition activity against HepG2 cell line (IC50 equals 3.2 mu g/mL) among all tested compounds. The results were compared to 5-Fluorouracil (5-FU) and doxorubicin as reference drugs, (IC50 5 and 3.56 mu g/mL). Furthermore, molecular docking of compounds 3b, 6a, and 8b into the binding site of topoisomerase II was carried out. The results of the binding energy scores of these compounds were compared to the docking score of Vosaroxin, a known 1,8-naphthyridine derivative which is in clinical trials as potential anticancer drug. Compound 8b docking result revealed that it is the only tested compound that intercalate with DNA segment of the topoisomerase II, similar to Vosaroxin which was used as reference drug for docking comparison.

The molecular energy changes accompanying the hydrogen migration and titration of H+ to different receptor-N at L-1 were calculated by density functional theory (DFT) at the SCRF-B3LYP/6-311++G(d,p) level in a CH2Cl2 solution, and the observed lowest-energy absorption and emission for L-1 and L-2 can be tentatively assigned to an intramolecular charge transfer (ICT) transition in nature.

For a specific fluorescent molecule, the increase of molecular conformation distortion is beneficial to endow it with aggregation-induced emission (AIE) and mechanofluorochromic (MFC) properties. Herein, 3,5-diphenyl-4H-pyran derivative5and 4,5-diphenyl-2,7-naphthidine derivative7with highly twisted conformations were synthesized. For compound5, although the introduction of phenyl rings with large steric hindrance at 3 and 5 positions of the 4H-pyran skeleton realized the transformation from aggregation-induced quenching (ACQ)-active molecule to AIE-active molecule, it only showed a low-contrast MFC activity. Compound7was accidentally obtained from compound5andn-butylamineviaa ring-opening and subsequent intramolecular ring-closing mechanism. Compound7was confirmed to have a highly twisted molecular conformation by the crystal structural analysis and exhibited AIE activity originated from the restriction of intramolecular rotation. Furthermore, compound7exhibited reversible high-contrast MFC activity. Upon grinding, the change of solid-state fluorescence color from orange to yellow was confirmed to be due to the partial destruction of crystal structure. This work provides new ideas for the design and synthesis of novel AIE-active and MFC-active fluorescent molecules based on ACQ-active parent molecules.
