Three novel Zn(II) complexes, [ZnCl2(qz)(2)] (1), [ZnCl2(1,5-naph)](n) (2) and [ZnCl2(4,7-phen)(2)] (3), where qz is quinazoline, 1,5-naph is 1,5-naphthyridine and 4,7-phen is 4,7-phenanthroline, were synthesized by the reactions of ZnCl2 and the corresponding N-heterocyclic ligand in 1:2 molar ratio in ethanol at ambient temperature. The characterization of these complexes was done by NMR, IR and UV-Vis spectroscopy, and their crystal structures were determined by single-crystal X-ray diffraction analysis. Complexes 1 and 3 are mononuclear species, in which Zn(II) ion is tetrahedrally coordinated by two nitrogen atoms belonging to two qz or 4,7-phen ligands, respectively, and by two chloride anions, while complex 2 is a 1D coordination polymer that contains 1,5-naph as bridging ligand between two metal ions. In agar disc-diffusion assay, complexes 1-3 manifested good inhibitory activity against two investigated Candida strains (C. albicans and C. parapsilosis), while not inducing toxic effects on the healthy human fibroblast cell line (MRC-5). This activity was not fungicidal, as revealed by the broth microdilution assay, however complex 3 showed the ability to modulate Candida hyphae formation, which is an important process during infection and showed significant synergistic effect with clinically used antifungal polyene nystatin.

A 2, 7-naphthyridine-based fluorescent turn-on probe for detection of biothiols in vitro and in vivo

A 2,7-naphthyridine derivative (AND-OH) was used as a fluorophore in the development of a novel fluorescent turn-on probe (AND-DNBS) by introducing a 2,4-dinitrobenzenesulfonate functionality. This fluorescent probe was successfully used for sensing glutathione (GSH) in aqueous media and showed favorable properties such as a rapid response time (45 s) and a significant Stokes shift (227 nm). The characteristics of the developed probe are better than those of another probe, namely ND-DNBS, which was also synthesized by our group and most other previously reported fluorescent probes for GSH. The AND-DNBS probe was able to detect GSH via a red emission in living A549 cells and zebrafishes. These results show that AND-DNBS has potential applications as an excellent probe for detecting GSH in various environments.

The chemistry of trinuclear silver(I) and copper(I) pyrazolates with aromatic, bidentate N-heterocycles, 2,2 ‘-bipyridine, 6,6 ‘-dimethyl-2,2 ‘-bipyridine, 1,10-phenanthroline, 2,9-dimethyl-1,10-phenanthroline, and 1,8-naphthyridine are presented. These N-donor ligands react readily with {[3,5-(CF3)(2)Pz]Ag}(3) and {[3,5-(CF3)(2)Pz]Cu}(3) providing mixed-ligand complexes with all nitrogen coordination spheres at metal in high yield. The fluorinated pyrazolate acts as a bridging ligand in these complexes. The bipyridine and phenanthroline systems with donor lone pairs at an acute angle serve as bidentate chelators for copper and silver providing dinuclear complexes. They feature planar, boat or chair shaped Ag2N4 or Cu2N4 cores and four-coordinate, tetrahedral metal sites. The 1,8-naphthyridine that has donor nitrogen lone pairs at a parallel orientation affords trinuclear copper and silver complexes with two- and three-coordinate metal centers. It uses only one of its lone pairs effectively in these adducts for metal ion coordination. In addition, 1,8-naphthyridine moieties of these copper and silver complexes show intermolecular pi-pi stacking. Results presented here show the effects of donor nitrogen lone-pair orientations of the bidentate N-heterocycles on the structures of the products resulting from their chemistry with {[3,5-(CF3)(2)Pz]Ag}(3) and {[3,5-(CF3)(2)Pz]Cu}(3).
