Lista et al., Nature Commun., 2017, 8, 16043). In this work, we synthesized and studied a series of 20 cationic bis(acylhy-drazone) derivatives designed as G4 ligands. The in vitro evaluation showed that most derivatives based on central pyridine (Py), naphthyridine (Naph) or phenanthroline (Phen) units were efficient G4 binders, in contrast to their pyrimidine (Pym) counterparts, which were poor G4 binders due to a significantly different molecular geometry. The influence of lateral heterocyclic units (N-substituted pyridinium or quinolinium residues) on G4-binding properties was also investigated. Two novel compounds, namely PyDH2 and PhenDH2, used at a 5 mu M concentration, were able to significantly enhance EBNA1 expression in H1299 cells in a GAr-dependent manner, while being significantly less toxic than the prototype drug PhenDC3 (GI(50) > 50 mu M). Antigen presentation, RNA pull-down and proximity ligation assays confirmed that the effect of both drugs was related to the disruption of NCL-EBNA1 mRNA interaction and the subsequent promotion of GAr-restricted antigen presentation. In this study, we describe the synthesis and biological effect of 2-oxo-1,8-naphthyridine-3-carboxamide derivative LV50, a new compound with high CB2 receptor (CB2R) affinity. We demonstrated that it decreases viability of Jurkat leukemia cells, evaluated by Trypan Blue and 3-(4,5dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), but mainly induces a proapoptotic effect. We observed an increase of a hypodiploid peak by propidium iodide staining and changes in nuclear morphology by Hoechst 33258. These data were confirmed by a significant increase of Annexin V staining, cleavage of the nuclear enzyme poly(ADP-ribose)-polymerase (PARP), and caspases activation. In addition, in order to exclude that LV50 non-specifically triggers death of all normal leukocytes, we tested the new compound on normal peripheral blood lymphocytes, excluding the idea of general cytotoxicity. To characterize the involvement of CB2R in the anti-proliferative and proapoptotic effect of LV50, cells were pretreated with a specific CB2R antagonist and the obtained data showed reverse results. Thus, we suggest a link between inhibition of cell survival and proapoptotic activity of the new compound that elicits this effect as selective CB2R agonist.

THE NEWLY synthesized compounds based on 2-amino-6-(2,4-dimethoxyphenyl)-4-(4-methoxyphenyl)nicotinonitrile 1. The reactivity of pyridine derivative 1 was studied towards different reagents by its reaction with malononitrile, 2-(4-chlorobenzylidene) malononitrile, ethyl cyanoacetate, cyano acetic acid, m-nitro benzaldehyde, sodium azide, formamide, acetic anhydride and/or acetic anhydride/H2SO4 to give compounds 2-5,7, 10-13, respectively. The reaction of 5 with acetyl acetone gave the bipyridine derivative 6, while reaction of compound 7 with chloroacetyl chloride and phenacyl bromide gave the azetidinone derivatives 8 and 9. The formimidate 14 were obtained via reaction of compound 1 with triethylorthoformate, while reaction of 14 with phenylene diamine and/or acetamide gave the formimidamide and pyridopyrimidine derivatives 15 and 16 respectively. Compound 1 was also allowed to react with urea, thiourea, phthalic anhydride, succinic anhydride, benzoyl chloride, chloroacetonitrile, chloroacetyl chloride, p-toluenesulfonylchloride and ethyl bromoacetate to give compounds 17a,b – 24. Reaction of compound 1 with dichloro reagents in 1:1 ratio gave the bicyclic derivatives 25a-c, while its reaction with oxalyl chloride, dichloro and tetrachlorobenzoquinone derivatives, and/or dichloronaphthoquinone gave the imidazopyridine derivatives 26-29. Reaction of compound 29 with o-phenylenediamine in 1:2 ratio afforded the di-condenesed product 30 while, reaction of compound 1 with dichlororeagents in 1:2 ratio gave the polyalkyl derivatives 31a-b. The newly synthesized compounds were characterized by IR, H-1-NMR, C-13-NMR and mass spectra. On the other hand the antimicrobial and anticancer activities of some of the newly synthesized compounds were studied and evaluated.

All compounds absorbed light >800 nm with maxima near 730 nm. Transient absorption (TA) measurements indicated thatn=4 thiophene rings (4T) positioned the PDT-active triplet intraligand charge transfer ((ILCT)-I-3) excited state in energetic proximity to the lowest-lying triplet metal-to-ligand charge transfer ((MLCT)-M-3).4Thad low-micromolar phototoxicity with PI(vis)and PI(733nm)values as large as 90 and 12, respectively. Spectroscopic studies suggested that the longer-lived (tau(TA)=3-6 mu s)(ILCT)-I-3 state was accessible from the(3)MLCT state, but energetically uphill in the overall photophysics. The study highlights that phototoxic effects can be achieved with NIR-absorbing Ru(II)PSs as long as the reactive(3)ILCT states are energetically accessible from the low-energy(3)MLCT states. It also demonstrates that tissue-penetrating NIR light can be used to activate the PSs in highly pigmented cells where melanin attenuates shorter wavelengths of light.
