In this article we disclose a series of new bisubstrate inhibitors of CK2 that are structurally represented by the oligo(L-Asp) peptide conjugates of benzo[c][2,6]naphthyridine-8-carboxylic acid. This fragment originated from CX-4945, the first in class inhibitor taken to clinical trials. The most potent conjugates possessed two-digit picomolar affinity and clear selectivity for CK2 alpha in a panel of 140 protein kinases. Labeling of the inhibitors with a fluorescent dye yielded probes for a fluorescence anisotropy-based binding/displacement assay which can be used for analysis of CK2 and precise determination of affinity of the highly potent (tight-binding) CK2-targeting inhibitors. New 1,8-naphthyridine-based probes for the selective fluorescence signalling of toxic cadmium: synthesis, photophysical studies and molecular modelling

Newly synthesised 1,8-naphthyridine-based molecular probes, NAP-1 and NAP-2, exhibit highly selective fluorescence responses towards the toxic cadmium over coordinatively competing Zn2+ and several other metal ions examined. On the one hand, NAP-1 (MeOH:H2O, v/v 80:20, pH 7.4) exhibits ca. 1.5 order of magnitude higher stability constant for Cd2+ over Zn2+; on the other hand, NAP-2 in MeOH offers unique selectivity only towards Cd2+, exhibiting both absorbance and emission red shifts as well as fluorescence enhancement. By 1H NMR analysis, the tetra-coordinated binding is indicated at least for the NAP-1+Cd2+ complex and theoretical calculations reveal relatively stronger binding of Cd2+ over Zn2+. [image omitted].

Identification and Characterization of a Selective Human Carbonyl Reductase 1 Substrate

During drug discovery efforts targeting inhibition of cytochrome P450 11B2 (CYP11B2)-mediated production of aldosterone as a therapeutic approach for the treatment of chronic kidney disease and hypertension, (S)-6-(5-fluoro-4-(1-hydroxyethyl)pyridin-3-yl)3,4-dihydro-1,8-naphthyridine-1(2H)-carboxamide (1) was identified as a potent and selective inhibitor of CYP11B2. Preclinical studies characterized 1 as low clearance in both in vitro test systems and in vivo in preclinical species. Despite low metabolic conversion, an active ketone metabolite (2) was identified from in vitro metabolite-identification studies. Due to the inhibitory activity of 2 against CYP11B2 as well as the potential for it to undergo reductive metabolism back to 1, the formation and elimination of 2 were characterized and are the focus of this manuscript. A series of in vitro investigations determined that 1 was slowly oxidized to 2 by cytochrome P450s 2D6, 3A4, and 3A5, followed by stereoselective reduction back to 1 and not its enantiomer (3). Importantly, reduction of 2 was mediated by an NADPH-dependent cytosolic enzyme. Studies with human cytosolic fractions from multiple tissues, selective inhibitors, and recombinantly expressed enzymes indicated that carbonyl reductase 1 (CBR1) is responsible for this transformation in humans. Carbonyl reduction is emerging as an important pathway for endogenous and xenobiotic metabolism. With a lack of selective substrates and inhibitors to enable characterization of the involvement of CBR1, 2 could be a useful probe to assess CBR1 activity in vitro in both subcellular fractions and in cell-based systems.

Two-photon absorption properties of BODIPY-like compounds based on BF2-naphthyridine complexes

Boron dipyrromethene type molecules (BODIPYs) are versatile molecules which have been used for applications ranging from photodynamic therapy to solar cells (DSSC). However, these molecules usually do not present high two-photon absorption cross-sections, limiting their use in nonlinear optical applications. Herein, we study a series of BF2-naphthyridine based boron-complexes with electron-donating and withdrawing groups to increase their two-photon absorption. We have found two-photon absorption cross-sections up to approximately 270 GM, which corresponds to an increase of approximately five times in comparison to the average cross-section value reported for molecules with similar conjugation length, indicating such compounds as potential materials for nonlinear applications in both the visible and infrared spectral regions.

Catalytic C(sp(2))-H Amination Reactions Using Dinickel Imides

C-H amination reactions are valuable transformations for the construction of C-N bonds. Due to their relatively high bond dissociation energies, C(sp(2))-H bonds are generally not susceptible toward direct nitrene insertion, necessitating alternative mechanisms for C-H activation. Here, we report that cationic dinuclear (NDI)Ni-2 (NDI = naphthyridine-diimine complexes catalyze intramolecular nitrene insertions into aryl and vinyl C(sp(2))-H bonds. Mechanistic studies suggest that a bridging imido ligand supported at a Ni-2 site induces C-H activation by a 1,2-addition pathway to generate an azametallacyclic intermediate. This organometallic mechanism contrasts with the electrocyclization/1,2-shift mechanism proposed for analogous transformations using Rh-2 catalysts. The implications of these mechanistic differences for the stereoselectivity and chemoselectivity of C-H amination are described.
