The mixture was purified by reverse phase ISCO (26 g C18 30 min gradient from 100percent A: 0percent B to 0percent A: 100percent B (A = 90percent H2O/10 percent ACN + 0.1percent TFA); (B = 90percent ACN/10percent H2O + 0.1percent TFA); detection at 220 nm) to yield E25B (18 mg, 5percent) as a minor byproduct. LCMS (ES): m/z 516.2 [M+H]+.

A stirred solution of 2-methyl-1,8-naphthyridine (57.5 g, 399 mmol) (available from Manchester Organics) and (R)-tert-butyl 3-(iodomethyl)pyrrolidine-1-carboxylate (124.2 g, 399mmcl) (Intermediate 1) in THF (1 L) was cooled to 0 ¡ãC and treated under nitrogen with a solution of lithium bis(trimethylsilyl)amide in THE (1M, 399 mL, 399 mmcl) over 20 mm and the reaction mixture was stirred at 0 ¡ãC for 3 h. The reaction was quenched with saturated ammonium chloride solution (500 mL) and water (500 mL) and ethyl acetate (1 L) was added. The layers were separated and the aqueous phase was extracted with further ethyl acetate (1 L). The combinedorganic layers were dried (MgSO4), filtered and evaporated in vacuo. The residual brown oil (162 g) was purified by chromatography on a silica cartridge (750 g) eluting with a gradient of 0 ? 100percent [ethyl acetate in (5percent MeOH ? 95percent ethyl acetate)] over 8 column volumes. The appropriate fractions were combined and evaporated in vacuo to give the title compound (46.65 g, 36percent) as an orange solid: LCMS (System A) RT=0.99 mm, 97percent, ES+ve m/z328 (M+H)?, [aiD2¡ã = + 22 (c 1.00in EtOH).

STEP 2. SYNTHESIS OF (E)-1-ETHOXY-2- (1, 8-NAPHTHYRIDIN-2- YL) ethanol. To the product from step 1, (81.5 G, 0.57 mol) in anhydrous THF (1.9 L) at – 40 C under Ar gas was added lithium bis (trimethylsilyl) amide (1 M in THF, 1.2 L, 1.2 MOL). After stirring for 30 min at-40 C, DIETHYLCARBONATE (72.5 mL, 0.60 mol) was added. The temperature of the reaction mixture was warmed up to 0 C and stirred for 2 h. The reaction mixture was quenched into saturated aq. NH4CI (700 mL) and the THF removed under reduced pressure. The resulting mixture was extracted with EtOAc (3 x 700 mL). The organic layers were combined, washed with brine, dried over NA2SO4, and concentrated under reduced pressure. The resulting residue was purified by flash column chromatography using 50percent EtOAc/hexane to give a yellow solid (81.2 g, 0.38 mol, 66percent).APOS;H NMR (400 MHz, DMSO-d6) 8 1.22 (t, 3H), 4.11 (q, 2H), 4.89 (s, 1H), 6.78 (d, 1H), 7.15 (dd, 1H), 7.47 (d, 1H), 7.80 (d, 1H), 8.36 (d, 1H), 11.8 (bs, 1H). a 2-Methyl-8-(tert-butoxycarbonyl)-5,6,7,8-tetrahydro-1,8-naphthyridine A mixture of 2-methyl-1,8-naphthyridine (J. Chem. Soc. (C) 1966, 315; 5.13 g, 35.58 mmole), 10percent Pd/C (1.14 g, 1.07 mmole), and absolute EtOH (70 mL) was deoxygenated through three evacuation/H2 purge cycles, then was stirred briskly under a balloon of H2. After 18.5 hr, the mixture was filtered through celite.(R)., and the filter pad was washed sequentially with absolute EtOH and EtOAc. The filtrate was concentrated to dryness, and the residue was reconcentrated from EtOAc to leave an off-white solid (5.25 g). A solution of the above material (5.25 g), di-tert-butyl dicarbonate (15.53 g, 71.16 mmole), and CH2Cl2 (10 mL) was concentrated on the rotavap to remove the solvent, and the oily residue was heated under N2 in an oil bath set at 55-60¡ã C. After 45 hr, the reaction was cooled to RT, and the residue was flash chromatographed on silica gel (40percent EtOAc/hexanes). The title compound (4.90 g, 55percent) was obtained as a light yellow solid: 1H NMR (300 MHz, CDCl3) delta7.27 (d, J=7.6 Hz, 1H), 6.81 (d, 7.6 Hz, 1H), 3.69 -3.79 (m, 2H), 2.65-2.75 (m, 2H), 2.48 (s, 3H), 1.83-1.98 (m, 2H), 1.52 (s, 9H), MS (ES) m/e 249 (M+H)+.

Five 20 mL microwave vials were each charged with 4-aminophenol (0.700 mg, 33 mmol) and 3 equivalents of cesium carbonate in 6.0 ml of DMF. The mixture was stirred at RT for 10 minutes. Following addition of 8-chloro-3-methoxy-1,5-naphthyridine (1 g, 26 mmol), the reaction vessels were capped and irradiated at 150 C. for 15 min in the microwave, at which time the reaction was determined complete by LCMS. The mixture was allowed to cool to ambient temperature and material from the five vessels was combined. A deep brown solid crashed out with addition of water. Filtered solids, washed with water and dried overnight in the vacuum oven to afford 4-(7-methoxy-1,5-naphthyridin-4-yloxy)benzenamine as a brown solid. [0359] Alternatively, the title compound may be prepared by the following method: In a nitrogen purged sealed pressure vessel, dissolved 4-aminophenol (0.617 g, 5.65 mmol) in DMF (0.030 L). Cesium carbonate (3.68 g, 11.3 mmol) was added and the mixture was stirred at RT for 5 min. Added 8-chloro-3-methoxy-1,5-naphthyridine (1.00 g, 5.14 mmol), heated to 90 C., stirred for 17 h. The mixture was allowed to cool to RT and was concentrated. The crude material was triturated with methanol, filtered, washed with methanol followed by water, and air dried to yield 4-(7-methoxy-1,5-naphthyridin-4-yloxy)benzenamine as brown solid. MS [M+H]=268.1; Calc’d 267.3 for C15H13N3O2.

A glass microwave reaction vessel was charged with 1-(trans-4-aminocyclohexyl)-3-cyclopropyl-1H-imidazo[4,5-b]pyrazin-2(3H)-one (0.1050 g, 0.339 mmol), 2-chloro-1,5-naphthyridine (0.067 g, 0.407 mmol, 00358), and diisopropylethylamine (0.177 ml, 1.017 mmol, Sigma-Aldrich Chemical Company, Inc.) in DMSO (0.484 ml) and heated to 120 C. for 2 days. The crude product was purified by reverse-phase preparative HPLC using a Phenomenex Gemini column, 10 micron, 100*50 mm, 0.1% TFA in CH3CN/H2O, gradient 10% to 90% over 15 min. Fractions containing product were collected and concentrated. The product was taken up in DCM and loaded onto a Silicycle Si-carbonate cartridge and washed with DCM and MeOH to remove any salts to give the title compound (6.4 mg, 0.016 mmol, 4.7% yield).
