Mechanistic evidence of a Gabriel Colman-type rearrangement

Rearrangement of quinolinimidoacetic acid derivatives induced by alkoxides leading to a mixture of 7-substituted 8-hydroxy-1,6-naphthyridin-5(6H)-ones and 6-substituted 5-hydroxy-1,7-naphthyridin-8(7H)-ones (Gabriel Colman-type reaction) was studied. Several chemical evidences accounts for a mechanism in two steps with quinolinamic ester intermediates (N-substituted 3-carbamoyl-2-pyridinecarboxylic acid alkyl esters and 2-carbamoyl-3-pyridinecarboxylic acid alkyl esters). These compounds were isolated from a reaction in mild conditions and their structure confirmed through an unequivocal synthesis.

A facile synthesis of highly fluorescent pyrido[2,3-d]pyrimidines and 1,8-naphthyridines via oxazine transformation and enaminic addition reactions

The synthesis and fluorescence properties of highly substituted bicyclic pryridine derivatives are described. 2-Substituted-4-oxo-pyrido[2,3-d]pyrimidines 2-8 resulted from the acylation reaction of alpha-aminonicotinonitrile 1 with aroyl chlorides, diethyl malonate, morpholine-4-carboxylate, acetic anhydride or formic acid under solvent-free conditions. [4+2] Cyclocondensation and cycloaddition reactions of compound type 1 with formamide and ammonium thiocyanate tolerated the fused pyrimidines 9 and 11, respectively. Finally, the Friedlander-like reaction was applied for the synthesis of 1,8-naphthyridines 12-15 via reaction of compound 1 with cyclohexanone, dimedone, acetyl acetone and benzyl methyl ketone, respectively, under AlCl3 catalysis. The fluorescence spectroscopic data of compounds 1-4, 6-8, 11, 12, 14 and 15 measured and significant results were observed.

1,8-Naphthyridinic fluorescent ‘turn-on’ and ‘turn-off’ chemosensors for detection of F- and Hg2+ ions mimicking INHIBIT molecular logic behaviour

Two new receptors viz. 2-bromoacetamido-5,7-dimethyl-1,8-naphthyridine (1a) and 2-chloroacetamido-5,7-dimethyl-1,8-naphthyridine (1b) containing 1,8-naphthyridine as fluorophore unit have been designed and synthesised. These receptors exhibited very high selectivity for F- ions among the various anions tested such as H2PO4-, OAc-, Cl-, Br-, I-, HSO4-, PF6-, ClO4-, BF4- and NO3- owing to anion-induced deprotonation of the amide NH by F- ions. Among the various cations tested (Na+, Ag+, Co2+, Ni2+, Cu2+, Zn2+, Cd2+, Hg2+, Pb2+ and Mn3+), 1a exhibited selective detection of Hg2+ ions by binding with N1 and N8 atoms of the naphthyridine moiety. Receptor 1a has been explored as a dual-ion detector for F- and Hg2+ ions in two contrasting modes (fluorescence ‘turn-on’ and ‘turn-off’) which is supported by spectroscopic and lifetime measurements. 1a exhibited lower detection limits for F- (0.4 ppm) and Hg2+ (2.4 ppm) ions. These receptors were recovered from anionic species of 1a formed after addition of F- ions by treating with protic solvents such as CH3OH. The mode of binding was also confirmed by spectroscopic and DFT studies. The chemical inputs of F- and Hg2+ ions generate an output which satisfies the condition of two-input INHIBIT logic operators.

New 1,4-dihydro[1,8]naphthyridine derivatives as DNA gyrase inhibitors

Owing to the growing need for novel antibacterial agents, we synthesized a novel series of fluoroquinolones including 7-substituted-1-(2,4-difluoropheny1)-6-fluoro-4-oxo-1,4-dihydro[1,8]naphthyridine-3-carboxylic acid derivatives, which were tested against clinically relevant Gram positive and Gram negative bacteria. Chemical structures of the synthesized compounds were identified using spectroscopic methods. In vitro antimicrobial effects of the compounds were determined via microdilution assay. Microbiological examination revealed that compounds 13 and 14 possess a good antibacterial profile. Compound 14 was the most active and showed an antibacterial profile comparable to that of the reference drugs trovafloxacin, moxifloxacin, and ciprofloxacin. A significant MIC90 value (1.95 mu g/mL) against S. aureus ATCC 25923, E. coli ATCC 35218, and E. coli ATCC 25922 was recorded for compound 14. We observed reduced metabolic activity associated with compounds 13 and 14 in the relevant bacteria via a luminescence ATP assay. Results of this assay supported the antibacterial potency of compounds 13 and 14. An E. coli DNA gyrase inhibitory assay indicated that compound 14 is a potent inhibitor of E. coli DNA gyrase. Docking studies revealed that there is a strong interaction between compound 14 and the E. coli DNA gyrase enzyme. Genotoxicity and cytotoxicity evaluations of compounds 13 and 14 showed that compound 14 is non-genotoxic and less cytotoxic compared to the reference drugs (trovafloxacin, moxifloxacin, and ciprofloxacin), which increases its biological importance. An unexpected semi-hydrogenation of a ligand in the complexation of 2,7-bispyridinyl-1,8-naphthyridine with Ru-3(CO)(12)

Thermal reaction of 2,7-bis(2-pyridinyl)-l, 8-naphthyridine (bpnp) with Ru-3(CO)(12) in the presence of moisture resulted in the formation of a formate-bridged diruthenium complex [(bpnp-H-3)Ru-2(mu-HCOO)(CO)(4)] (1), in which the ligand was partially hydrogenated. Complex 1 was fully characterized by spectroscopic analyses and X-ray single crystal determination. Regarding the partially reduced ligand in 1, it occurs through a water-gas shift type reduction. The bridging formate ligand can be substituted by other carboxylate ligands. Physical and chemical properties of the newly prepared complexes were investigated.

Preliminary Investigation of a Novel F-18 Radiopharmaceutical for Imaging CB2 Receptors in a SOD Mouse Model

We successfully radiolabelled a novel prospective cannabinoid type 2 receptor ligand with F-18 and tested its biodistribution in animal models by positron emission tomography (PET)/computed tomography (CT) imaging. The radiolabelling process was conducted on an alkyl mesylate fragment of the main naphthyridine core, using highly efficient microfluidic technology. No preliminary protection was needed, and the product was purified by semi-prep HPLC and SPE formulation, allowing the desired diastereomeric mixture to be obtained in 29% radiochemical yield and >95% radiochemically pure.
