General procedure: To an indole-3-acetonitrile derivative (1.0 equiv) dissolved in anhydrous methanol (4mL for 2.31mmol of starting material) in a dried microwave vial, sodium methoxide (1.7 equiv) was added and stirred at room temperature for 15min protected from light. Quinoline/isoquinoline-carboxaldehyde derivative (1.2 equiv) was added and the mixture was subjected to microwave irradiation at 95C for 8.5min. The reaction was cooled to room temperature and then chilled in an ice/salt bath. The resulting precipitate was filtered, washed with methanol, and dried under vacuum to afford a solid as the product.

To 5,7-dichloro-1 ,6-naphthyridine (5.01 g, 25.2mmol) and 1 , 1 -dimethylethyl (3R)-3- (aminomethyl)-3-fluoro-1 -piperidinecarboxylate (5.32g, 22.90mmol) in NMP (20ml) was added DI PEA (8.00ml, 45.8mmol) and the mixture was stirred at 100C for 72h under nitrogen. The reaction was cooled and partitioned between ethyl acetate and water (200ml each). The aqueous was washed with ethyl acetate. The combined organics were washed with water and the solvent was removed. The residue was dissolved in DCM and loaded onto a 100g silica SNAP column and purified on the SP4 eluting with 0-50% ethyl acetate in cyclohexane over 17CVs. Appropriate fractions were combined and the solvent was removed to give the title compound as a pale orange solid which was dried under high vacuum for 2h (7.61 g).LCMS (Method B): Rt = 1 .12min, MH+ 395/397

To this was added N1-(4-phenylphthalazin-1-yl)benzene-1,4-diamine (124 mg, 396 mumol) and the reaction mixture was stirred at 70 C. for 17 h. Upon cooling to RT, the mixture was dissolved in DMF and purified using Gilson reverse phase chromatography. The product fractions were combined, concentrated and the resulting crude was extracted into DCM, washed 1* sodium carbonate, 1*H2O, dried with Na2SO4, filtered through fritted funnel, concentrated to yield N1-(7-methoxy-1,5-naphthyridin-4-yl)-N4-(4-phenylphthalazin-1-yl)benzene-1,4-diamine as light yellow solid. Under Ar(g), to a mixture of pyrazin-2-amine (1) (209mg, 2.2mmol), 2- chloro-1 ,5-naphthyridine (2) (329mg, 2.0mmol), Cs2C03 (1.30g, 4.0mmol) was added degassed dry 1 ,4-dioxane (13ml_). The reaction mixture was then flushed with Ar(g) for 1 min before Pd2(dba)3 (92mg, 0.1 mmol) and Xantphos (127mg, 0.22mmol) were added. The reaction mixture was heated up to 90C for 40h. It was then cooled down to rt and concentrated in vacuo, CH2CI2 (15ml_) and H20 (15ml_) were added. The organic phase was separated and the water layer was extracted with EtOAc (15ml_). The organic layers were combined and Pd- scavenger (MP-TMT, ~400mg, 1.3mmol/g) was added. This was shaken for several hours followed by filtration. The filtrate was concentrated in vacuo, dissolved in DMSO (4ml_) and purified by basic prep LCMS to yield (3) as a solid (121 mg, 27%). (0160) LCMS (ES): Found 224.3 [M+Hf.

KMnO4 (96 mmol, 15.2 g, purchased from Beijing Chemical Industry Plant) was added at room temperature. After stirring for 6 h at 25¡ã C., the mixture was suction filtered, and the filtrate was evaporated under reduced pressure to obtain a crude solid product, which was further purified by column chromatography (petroleum ether/ethyl acetate mixture with a volume ratio of 100/1). Method A B-9 Ex. 21 To a stirred solution of Intermediate B-9 (99.0 mg, 0.231 mmol) and 8-chloro-3-methoxy- 1 ,5-naphthyridine (30.0 mg, 0.154 mmol) in THF (4 ml) under nitrogen was added chloro[2- (dicyclohexylphosphino)-3,6-dimethoxy-2′,4′,6′-triisopropyl-l ,l ‘-biphenyl][2-(2- aminoethyl)phenyl]palladium(II) (24.6 mg, 0.0310 mmol) and sodium teri-butoxide (0.154 mL, 2 M) at RT. The mixture was stirred at 40 C for 15 h, cooled, filtered, and diluted with water (5 ml) and extracted with EtOAc (6 mL x 3). The organic layers were collected, dried with sodium sulfate, filtered, and concentrated. The residue was dissolved in DCM (6 mL), treated with TFA (0.200 mL) and stirred at 18 C for 1 h. The mixture was quenched with aqueous sodium hydrogen carbonate (5 ml) and extracted with EtOAc (5 mL x 4). The combined organic extracts were washed with brine, dried (Na2S04), filtered and concentrated. The residue was purified by prep-HPLC (column 250 x 21.2 mm, 4 muiotaeta; mobile phases A = 0.075 % TFA water, B = MeCN; gradient 10-40% B, 11 min, 25 mL/min) to provide example 21. MS for example 21: m/e = 485 (M+l).
